A Study Proves NNB MitoPrime® (L-Ergothioneine) Exhibits a strong protective effect against UVA-mediated mtDNA deletion which might smooth and brighten people’s skin.

Wrinkles, roughness and pigmentation are all inevitable skin issues we pay a lot attention to as we age. A lot of us naturally put the blame on getting another year older. Little imagined the culprit of all that is Photoaging (also known as “Sun Damage”), premature aging of the skin caused by repeated exposure to ultraviolet radiation (UV), primarily not only from the sun but also from artificial UV sources. The UV radiation that affects the skin is composed of two different types of waves, UVA and UVB.
When UV rays hit the skin, they damage its DNA, and cells in the dermis scramble to produce melanin in the epidermis to prevent further damage. One of the most common types of damage of mitochondrial DNA (mtDNA) is the so-called “common deletion (Cdel)” referring to a deletion of 4977 base pairs (bp). Intra individual comparison studies have revealed that the “common deletion” is increased up to 10-fold in photoaged skin, as compared with sun-protected skin of the same individuals. In the skin cells, this phenomenon is probably caused by oxidative damage of mtDNA induced by UV and is involved in skin photoaging, characterized by wrinkles, uneven pigmentation, brown spots, laxity, and a leathery appearance.
In the latest study, we evaluated a UV-mediated mitochondrial DNA deletion in normal human dermal fibroblasts (NHDF) under UV-irradiated conditions. Briefly, NHDF pre-treated with the test compound(ergothioneine/ resveratrol), were exposed 3 times a day to a UVA irradiation (in absence of the test compound), followed by a treatment renewal. This protocol was repeated almost every day (except for the weekend) for 16 days. The relative quantification of the Cdel quantity was carried out by using quantitative PCR (qPCR) method.
Below is the Effect of NNB L-Ergothioneine and potential reference resveratrol on the UV-induced mtDNA common deletion (Cdel) in fibroblasts we found in this study:
As can be seen from the PCR electrophoresis band, L-Ergothioneine significantly prevents the deletion leading to wrinkles and damaged skin. It is worth nothing that L-Ergothioneine may have better protection effect than that of resveratrol.
This study indicates that L-Ergothioneine might be involved in the protection of mtDNA from ROS generated in the course of electron transport. “That is to say, L-Ergothioneine may act as a promising anti-photo-aging ingredient which is a huge opportunity for skin care company to put this novel ingredient in their UV protection products.” says Dr. Fan, director of clinical department of NNB Nutrition.
